Resumen

La estandarización de técnicas moleculares como la reacción en cadena de la polimerasa para la amplificación de la región código de barras constituye un paso crucial en el laboratorio hacia la implementación de esta estrategia en moscas de importancia forense. Tradicionalmente, la búsqueda de condiciones óptimas requiere la ejecución de un gran número de experimentos independientes que varían en las concentraciones de los componentes de la reacción. En este estudio se implementó la metodología Taguchi para establecer las condiciones óptimas usando un número mínimo de experimentos en moscas de la familia Calliphoridae. La cantidad de ADN, así como la temperatura de alineamiento de los cebadores representaron los factores más relevantes en el éxito de la amplificación. Esta metodología puede aplicarse para la estandarización de otras técnicas moleculares como una estrategia simple, rápida y de bajo costo.
